PromethION 2 Solo Quick Start Guide

This guide provides essential information for setting up your PromethION 2 Solo and ensuring it is ready for use.

The PromethION 2 Solo features the following components:

Installation Guidance

To use the device with a laptop:

  1. Ensure your computer meets the IT requirements.
  2. Power on your laptop.
  3. Install the latest MinKNOW software for MinION Mk1B and PromethION 2 Solo devices from the Software Downloads page (community.nanoporetech.com/downloads). Click the Nanopore wheel icon on the desktop after installation.

To use the device with GridION:

  1. Check that the GridION serial number on the label is GXB002 or above.
  2. Power on your GridION.
  3. Update to the latest GridION software from the Software Downloads page (community.nanoporetech.com/downloads). Click the Nanopore wheel icon on the desktop after updating.

Device Setup

  1. Plug the AC power adapter into the mains supply and the other end into the PromethION 2 Solo power port.
  2. Ensure no Configuration Test Cells (CTCs) are inserted into the PromethION 2 Solo.
  3. Plug the USB Type-C cable into the correct ports on your host laptop or GridION and the other end into the PromethION 2 Solo, with the P2 icon on both USB Type-C connectors facing upwards.
  4. Toggle the power switch on the PromethION 2 Solo to the 'On' position.

Perform a Hardware Check

A hardware check is required before performing your first PromethION 2 Solo sequencing run. You will need your CTCs for this procedure.

Hardware Check Overview:

  1. Load both CTCs into the PromethION 2 Solo Flow Cell positions and close the device lid.
  2. Wait until the flow cell positions initialize with a blue LED. In MinKNOW software, the flow cell status indicators (two boxes) will change from grey to white. (Note: LEDs pulse white then turn solid blue when CTCs are inserted).
  3. Press 'Select all available positions'. The flow cell status indicators will change to dark blue. (Note: LEDs pulse green during the hardware check).
  4. Press 'Start' in the bottom right of the MinKNOW display.
  5. Check for a ✔️ symbol above the flow cell icon in the flow cell positions to indicate a passed hardware check.
  6. Remove the CTCs from the flow cell positions after completing the hardware check.

If your hardware check fails, refer to 'Support' in the Additional Information section.

Powering Off the Device

To power off your PromethION 2 Solo:

  1. Remove the PromethION CTCs from the device.
  2. Toggle the power switch to the 'Off' position.

To power off your laptop or GridION:

To power off your GridION using MinKNOW, follow the workflow in the software (Host settings > Shutdown).

You can then close MinKNOW and power off your laptop.
