1. Introduction
The MAGUS Lum 400 Fluorescence Microscope is a sophisticated instrument designed for advanced diagnostic and research applications. It primarily utilizes fluorescence (reflected light) and brightfield (transmitted light) microscopy techniques. The microscope's robust design and versatile features allow for detailed observation and analysis in various scientific fields, including DNA analysis, pathogen study, sanitary and epidemiological surveillance, and forensic examination. With optional equipment, it can also perform darkfield, phase contrast, and polarization methods.
This manual provides essential information for the safe and effective use of your MAGUS Lum 400 microscope, covering setup, operation, maintenance, and troubleshooting.
2. Safety Information
Please read and understand all safety precautions before operating the microscope. Failure to do so may result in injury or damage to the instrument.
- Electrical Safety: Ensure the microscope is connected to a grounded power outlet with the correct voltage (85–265V, 50/60Hz). Do not operate with wet hands. Disconnect power before cleaning or servicing.
- Mercury Lamp Safety: The reflected light source uses a 100W mercury lamp. Mercury lamps operate at high pressure and temperature. Always allow the lamp to cool down completely before handling. Avoid direct eye exposure to the mercury lamp's UV light. Use appropriate protective eyewear if servicing the lamp.
- Optical Safety: Never look directly into the light source. Use caution when handling objectives and eyepieces to avoid scratching optical surfaces.
- Handling: Always carry the microscope by its main body, supporting the base. Avoid sudden impacts or vibrations.
- Ventilation: Ensure adequate ventilation around the microscope, especially near the lamp housing, to prevent overheating.

4. Setup
Follow these steps to set up your MAGUS Lum 400 microscope:
- Unpacking: Carefully remove all components from the packaging. Retain the original packaging for future transport or storage.
- Placement: Place the microscope on a stable, level surface, away from direct sunlight, vibrations, and excessive dust. Ensure sufficient space for operation and ventilation.
- Head Installation: Gently insert the trinocular head into the top port of the microscope body. Secure it with the locking screw.
- Eyepiece Installation: Insert the eyepieces into the eyepiece tubes of the trinocular head.
- Objective Installation: Screw the objectives into the revolving nosepiece in ascending order of magnification (e.g., 4x, 10x, 40x, 100x).
- Lamp Installation (if not pre-installed): Refer to the specific instructions for installing the 100W mercury lamp and 30W halogen lamp. Ensure proper alignment and secure fastening.
- Power Connection: Connect the power cord to the microscope's power input and then to a suitable grounded electrical outlet.

5. Operating Instructions
This section outlines the basic operation of the MAGUS Lum 400 microscope.
5.1. Power On and Illumination
- Turn on the main power switch.
- For transmitted light (brightfield), activate the 30W halogen lamp. Adjust the brightness using the intensity control knob.
- For reflected light (fluorescence), activate the 100W mercury lamp. Allow a warm-up period as specified by the lamp manufacturer. Ensure appropriate filter cubes are in place for the desired excitation and emission wavelengths.
- Köhler Illumination: For optimal image quality in both transmitted and reflected light, set up Köhler illumination. This involves adjusting the field diaphragm and aperture diaphragm to achieve even and bright illumination across the field of view.
5.2. Specimen Placement and Focusing
- Place your specimen slide on the mechanical stage, securing it with the stage clips.
- Rotate the revolving nosepiece to select the lowest magnification objective (e.g., 4x).
- Using the coarse focus knob, raise the stage until the objective is close to the specimen, then slowly lower it while looking through the eyepieces until the image comes into rough focus.
- Use the fine focus knob for precise focusing.
- Adjust the interpupillary distance and diopter settings on the eyepieces for comfortable viewing.
5.3. Magnification and Observation
- The microscope offers a magnification range of 40x to 1000x, depending on the objective and eyepiece combination.
- To increase magnification, rotate the nosepiece to a higher power objective. Re-focus using the fine focus knob.
- When using the 100x objective, apply a small drop of immersion oil to the specimen slide before lowering the objective into the oil. After use, clean the oil immersion objective thoroughly.
- For fluorescence observation, ensure the correct filter cube is engaged and the mercury lamp is stable. Observe the fluorescent signal against a dark background.
6. Maintenance
Proper maintenance ensures the longevity and optimal performance of your microscope.
- Cleaning Optics: Use only specialized lens cleaning solution and lens paper or a soft, lint-free cloth to clean objective lenses and eyepieces. Breathe lightly on the lens surface before wiping to help remove dust. Never use abrasive materials.
- General Cleaning: Wipe the microscope body with a soft, damp cloth. Avoid using organic solvents.
- Dust Protection: Always cover the microscope with the provided dust cover when not in use to prevent dust accumulation.
- Lamp Replacement: Replace lamps according to manufacturer guidelines or when their performance degrades. Always disconnect power and allow lamps to cool before replacement.
- Storage: Store the microscope in a dry, dust-free environment. If storing for extended periods, remove eyepieces and objectives and store them in their protective containers.
7. Troubleshooting
This section addresses common issues you might encounter.
| Problem | Possible Cause | Solution |
|---|---|---|
| No image or very dim image | Power off; lamp burnt out; aperture/field diaphragm closed; objective not clicked into place. | Check power connection and switch; replace lamp; open diaphragms; rotate nosepiece until objective clicks. |
| Image blurry or out of focus | Incorrect focus adjustment; objective/eyepiece dirty; slide upside down. | Adjust fine focus; clean optics; reorient slide. |
| Dark spots in field of view | Dust on eyepiece, objective, or condenser. | Clean optics. Rotate eyepiece to determine if dust is on it. |
| Uneven illumination | Köhler illumination not set correctly; lamp misaligned. | Re-adjust Köhler illumination; check lamp alignment. |
| No fluorescence signal | Mercury lamp off or burnt out; incorrect filter cube; specimen not fluorescent. | Check mercury lamp status; select correct filter cube; verify specimen preparation. |
