1. Introduction

This manual provides essential information for the safe and efficient operation of the Hettich Mikro 200 Microliter Centrifuge, Model 200MICRO24. This instrument is designed for research centrifugation tasks, particularly for DNA, RNA, and other temperature-sensitive samples.

2. Safety Information

  • Always balance tubes precisely before centrifugation to prevent vibration and damage to the unit.
  • Never open the lid while the rotor is spinning. The safety-lock lid prevents this during operation.

  • Wear appropriate personal protective equipment (PPE), such as lab coats, gloves, and eye protection, when handling samples.
  • In case of a spill or breakage, follow laboratory safety protocols for decontamination.
  • Disconnect power before cleaning or maintenance.

3. Setup

Follow these steps to set up your Hettich Mikro 200 centrifuge:

  1. Unpacking: Carefully remove the centrifuge and all accessories from the packaging. Retain packaging for future transport or storage.
  2. Placement: Place the centrifuge on a sturdy, level laboratory bench. Ensure there is adequate clearance (at least 10 cm) around the unit for ventilation.
  3. Power Connection: Connect the power cord to the centrifuge's power inlet and then to a grounded 115V AC power outlet.
  4. Rotor Installation: Ensure the rotor is correctly seated on the motor shaft. The Mikro 200 is equipped with a 24-place rotor for 1.5-2.0 mL tubes.

4. Operating Instructions

Operating the Hettich Mikro 200 is straightforward. Follow these steps for typical centrifugation runs:

  1. Power On: Turn on the main power switch, usually located at the rear or side of the unit. The display will illuminate.
  2. Load Samples: Open the centrifuge lid using the OPEN button. Load your samples into the rotor, ensuring that tubes of equal weight are placed opposite each other for proper balancing. The rotor accommodates 24 tubes of 1.5-2.0 mL capacity.
  3. Close Lid: Close the centrifuge lid firmly until it latches securely.
  4. Set Parameters:
    • Use the SELECT button to toggle between setting RPM (revolutions per minute) or RCF (relative centrifugal force) and time.
    • Adjust the values using the rotary knob. The display shows current settings, for example, "14000 RPM 10:00 min".
    • If refrigeration is needed, the unit's fast cool function can bring the temperature down to +4 °C in 10-15 minutes.
  5. Start Run: Press the START button to begin the centrifugation cycle. The lid will lock automatically.
  6. Monitor Progress: The display will show the remaining time and current speed.
  7. End of Run: Once the cycle is complete, the rotor will come to a complete stop, and the lid will unlock automatically.
  8. Unload Samples: Press the OPEN button to open the lid and carefully remove your samples.
  9. Power Off: Turn off the main power switch when the centrifuge is not in use.

5. Maintenance

Regular maintenance ensures the longevity and optimal performance of your centrifuge.

  • Cleaning:
    • Wipe down the exterior of the centrifuge with a damp cloth and mild detergent. Avoid abrasive cleaners.
    • Clean the rotor and rotor chamber regularly. Ensure all spills are immediately cleaned to prevent corrosion.
    • The bio-containment, autoclavable, phenol-resistant lids should be cleaned according to laboratory protocols.
  • Rotor Care: Inspect the rotor for any signs of damage, cracks, or corrosion before each use. Do not use a damaged rotor.
  • Lid Seal: Check the lid seal for any damage or wear. A damaged seal can affect performance and safety.
  • Ventilation: Ensure the ventilation openings are clear of dust and debris to prevent overheating.
