Hygiena UltraSnap Surface ATP Test Reagent Instructions
Product: UltraSnap® Surface ATP Test Reagent
Product Number: US2020 (100 Tests)
Brand: Hygiena
Certification: AOAC RESEARCH INSTITUTE LICENSE NUMBER 101803
Product Overview
The UltraSnap® Surface ATP Test Reagent is a self-contained device designed for use with Hygiena® luminometers. This system monitors surface hygiene in a wide range of industries by measuring Adenosine Triphosphate (ATP). ATP is a universal energy molecule found in all animal, plant, bacterial, yeast, and mold cells. Organic residues on surfaces contain ATP, and microbial contamination also contains ATP, though typically in lower amounts. Proper cleaning should significantly reduce all ATP sources.
When a sample is collected and comes into contact with the unique liquid-stabilized luciferase/luciferin reagent in the UltraSnap device, the emitted light intensity is directly proportional to the amount of ATP present in the sample. The luminometer measures this light intensity and reports the result in Relative Light Units (RLU). Higher RLU values indicate higher ATP content and dirtier surfaces.
Intended User
Laboratory personnel with standard laboratory operation training are qualified to use the UltraSnap device.
Applicability
The UltraSnap device is suitable for measuring ATP from environmental surfaces. This method has been validated by the AOAC Research Institute Performance Tested Method™ (PTM) program and is applicable to major food categories including meat, dairy, and beverages. For details, refer to AOAC RI PTM Certificate 101803 at www.hygiena.com/documents.
Limitations
The UltraSnap test is designed to detect invisible or trace amounts of residue. Swabbing visibly soiled surfaces with the swab may result in excessive soil load, which can inhibit the bioluminescence reaction and produce inaccurate results.
For water sample testing, such as Clean-In-Place (CIP) rinse water, use the AquaSnap® Water ATP testing device (Product Numbers: AQ-100X and AQ-100FX). For more information, visit www.hygiena.com.
Important Notes and Precautions
- Before use, ensure the UltraSnap test device has equilibrated to room temperature (21-25°C / 70-77°F).
- The swab tip is pre-moistened to ensure maximum sample collection.
- Condensation may be present in the swab tube; this is normal.
- Turn on the luminometer. If the luminometer has a pre-set test position, select the appropriate test position before starting the test.
Test Procedure
- Firmly hold the swab tube, twist and pull out the swab tip to detach it from the tube.
- Thoroughly swab a standard 10x10 cm (4x4 inch) area on typical flat surfaces. Important swabbing technique tips:
- For irregular surfaces, maintain a consistent swabbing technique for each test; swab a sufficiently large area to collect representative samples.
- Do not touch the swab or the inside of the sample collection device with your fingers.
- Swab in a cross-pattern, vertically, horizontally, and diagonally in both directions.
- Rotate the swab during sample collection to ensure the swab tip fully collects the sample.
- Apply enough pressure to bend the swab shaft.
- Return the swab to the swab tube.
- To activate the device, firmly hold the swab tube, bend the Snap-Valve forward and backward with your thumb and forefinger to break the valve. Squeeze the bulb twice to expel all liquid into the swab tube.
- Immerse the swab tip in the liquid for 5-10 seconds. After activation, the sample must be read in the luminometer within 30 seconds.
- Hold the luminometer vertically and insert the entire UltraSnap device into the Hygiena luminometer.
- Refer to the instrument manual for operating instructions. Brief steps:
- a. If using an EnSURE® Touch luminometer, close the lid and press the 'Run Test' button to initiate measurement. Results will be displayed in 10 seconds.
- b. If using an EnSURE® or SystemSURE Plus® luminometer, close the lid and press the 'OK' button to initiate measurement. Results will be displayed in 15 seconds.
Diagram Description: The procedure involves preparing the swab, swabbing a 10x10 cm area with specific techniques (cross-pattern, rotation, pressure), returning the swab to the tube, activating the device by breaking the Snap-Valve and squeezing the bulb to mix the sample with the reagent, immersing the swab tip, and inserting the device into the luminometer for measurement. The final step shows a smartphone displaying test results.
Additional Information
Results Interpretation
Hygiena luminometers are pre-set with pass/fail RLU limits (Table 1), based on industry standards and published research.
Table 1. Hygiena Luminometer Default RLU Limit Settings.
Interpretation | EnSURE Touch (RLUs) | EnSURE or SystemSURE Plus (RLUs) |
---|---|---|
Pass (Clean) | ≤20 | ≤10 |
Caution* (Warning) | 21-59 | 11-29 |
Fail (Contaminated) | ≥60 | ≥30 |
*Insufficient cleaning.
Hygiena recommends setting RLU thresholds based on your facility's standards. For guidance, consult the technical bulletin "Lower and Upper RLU Limits for ATP Monitoring Programs". For optimal system utilization, use SureTrend® software to track and analyze test performance over time.
