6. Tubing Operation Instructions

This system has multiple operating stages. Please refer to the following instructions for setting up the tubing. For cassette-related cleaning steps and usage of solutions, please refer to the operation manuals of each brand.

(1) Cleaning

A diagram illustrates the tubing setup for cleaning.

a. For initial operation, the entire system must be cleaned. Connect the cleaning tube between the return pressure manifold (upper) and the inlet pressure manifold (lower), as shown in Figure 1.

b. Open the concentration bottle cap, pour in the cleaning solution, and close the bottle cap.

c. Refer to Figure 1 for the three-way valve and flow path settings.

d. Start the peristaltic pump and slowly increase the speed to 50 rpm.

e. Allow the system to circulate for 15-30 minutes, then stop the peristaltic pump.

(2) Drainage and Sample Collection

Diagrams illustrate the tubing configurations for drainage and sample collection.

a. With the peristaltic pump stopped, set the three-way valve direction as shown in Figure 2.

b. Open one of the airtight caps on the concentration bottle. The liquid in the concentration bottle will be collected into a container via the collection tubing due to gravity.

c. If the system tubing needs to be drained after the experiment, stop the peristaltic pump, adjust the three-way valve direction, remove the filtration cassette, and connect the cleaning tube, as shown in Figure 3.

d. As shown in Figure 4, adjust the three-way valve direction and start the peristaltic pump to collect all the solution in the system tubing into the concentration bottle, then stop the peristaltic pump.

e. As shown in Figure 5, adjust the three-way valve at the bottom of the concentration bottle, open one of the airtight caps on the concentration bottle, and the solution will naturally drain out through the collection tubing by gravity.

f. If gravity drainage is insufficient, adjust the three-way valve and flow path as shown in Figure 6, start the peristaltic pump, and drain the solution from the drain tubing using the pump's power.

(3) Cassette Rinsing

A diagram shows the tubing setup for cassette rinsing.

a. Adjust the three-way valve direction and flow path as shown in Figure 7. Pour the rinsing solution into the concentration bottle.

b. Start the peristaltic pump and slowly increase the speed until the inlet pressure reaches the recommended pressure for the cassette.

Note: Check the tubing for leaks. If leaks are found, tighten the connectors.

c. Continuously circulate the rinsing solution. Refer to the operation manual of each cassette brand for the recommended rinsing solution volume.

II. Cassette and System Rinsing

a. Adjust the three-way valve direction and flow path as shown in Figure 8.

b. Start the peristaltic pump and slowly increase the speed until the inlet pressure reaches the recommended pressure for the cassette.

Note: Check the tubing for leaks. If leaks are found, tighten the connectors.

c. Adjust the pressure regulating clamp and peristaltic pump speed until the return and inlet pressures reach the recommended pressures for the cassette.

d. Continue circulating until the concentration reaches the desired level. Stop the peristaltic pump and refer to (2) Drainage Steps to drain the solution from the tubing into a container.

(4) Pre-treatment and Concentration

I. Pre-treatment

a. Adjust the three-way valve direction and flow path as shown in Figure 9.

b. Pour the buffer solution into the concentration bottle, start the peristaltic pump, and slowly increase the speed. Refer to the recommended cassette values for inlet pressure, and ensure the tubing is filled with solution to avoid air bubbles.

Note: Check the tubing for leaks. If leaks are found, tighten the connectors.

c. When the liquid level in the concentration bottle reaches the bottom, stop the peristaltic pump. Remove the filtration tubing, pour out the liquid in the tubing, and reinstall the cassette to complete the pre-treatment before the concentration experiment.

II. Concentration

a. Pour the sample into the concentration bottle, start the peristaltic pump, and refer to the recommended cassette values for inlet pressure.

b. Adjust the pressure regulating clamp and peristaltic pump speed until the inlet and return pressures reach the recommended cassette values.

Note: Ensure the inlet pressure does not exceed the recommended cassette value, and the return pressure does not exceed 40 psi.

c. Continue circulating until the desired concentration is reached, then stop the peristaltic pump.

d. Refer to (2) Drainage Steps to collect the sample from the system into a container.

(5) Continuous Sampling

If the sample volume to be filtered exceeds the concentration bottle capacity of 500 mL, the excess sample can be placed in another beaker. Use a silicone tube, with one end inserted into the septum on the concentration bottle cap and the other end placed in the beaker. As shown in Figure 10. During the filtration process, the concentration bottle will gradually become negative pressure, drawing the sample from the beaker into the concentration bottle for continuous sampling.
