Brightfield Observations in Transmitted Light
Switching on the Illumination: Ensure correct input voltage. Turn the ON/OFF switch to the 'ON' position (marked '–'). Adjust brightness to approximately 70%. Avoid prolonged use at maximum brightness.
Placing the Specimen: Place the specimen in the center of the stage. The stage has XY control knobs for movement (75mm X-axis, 50mm Y-axis).
Using the Beam Splitting: The beam splitter lever controls the light path. Position it for eyepiece observation (symbol ) or camera port (symbol ).
Focusing on the Specimen: Use coarse and fine focusing knobs. Start with the 10x objective. Raise the stage with the coarse knob, then slowly lower it while looking through the eyepiece until the image appears. Use the fine knob for sharp focus. Engage the coarse focusing lock for high magnification. Adjust coarse focusing tension as needed.
Adjusting the Eyepiece Tubes: Compensate for ametropia by adjusting the diopter ring on the left eyepiece. Focus first with the right eyepiece (left eye closed), then with the left eyepiece (right eye closed) using the diopter ring.
Setting up Köhler Illumination: This technique optimizes image quality, resolution, and even illumination. Steps involve adjusting the power, transmitted light switch, beam splitter, specimen placement, condenser height, field diaphragm, and aperture diaphragm. The goal is to achieve sharp, evenly lit images with minimal artifacts.
Calculating Magnification and Field of View: Total magnification is eyepiece power multiplied by objective power. Field of view is calculated by dividing the eyepiece field number by the objective magnification. A stage micrometer is used for precise measurements.
Diagrams (Fig. 6-12) illustrate these procedures.
Using the Digital Camera
The digital camera captures 4K images via HDMI/USB 3.0 or Full HD via Wi-Fi. It mounts to the trinocular tube. Ensure proper camera selection for specific tasks. To enable the camera:
- Loosen the dust cap screw and remove the dust cap.
- Connect the camera to the C-mount adapter.
- Fit the camera into the trinocular tube and secure it.
- Set the beam splitter lever to the camera position.
- Switch on the camera and adjust focus using the fine focusing knob.
- For synchronized imaging, use the trinocular tube's centering screws to align the camera image with the eyepiece view.
Diagrams (Fig. 13-14) illustrate camera setup and image adjustment.
Using the Monitor
The monitor displays real-time images from the camera. Connect the monitor to the camera using an HDMI cable and connect both to power. Switch on the monitor and adjust focus as needed.
Diagram (Fig. 15) shows monitor connection.
Reflected Light Observations
General Procedure: Switch on illumination, place specimen, check trinocular tube, and focus. Set the lever to the reflected light position ( ). Switch on the reflected light illuminator.
Centering the Light Source: If re-centering is needed after transport, place white paper on the stage, introduce the free slot of the revolving nosepiece, open diaphragms, and adjust the collector and alignment knobs to center the filament image.
Setting up Köhler Illumination (Reflected Light): Ensure power and illuminator are on. Introduce the 10x objective, place the specimen, and focus. Close field and aperture diaphragms to illuminate the center of the field of view. Verify diaphragm centering. Open the field diaphragm until it just disappears from view and close the aperture diaphragm for optimal contrast and illumination. Adjust diaphragms when changing objectives.
Using Color Filters: A slider with color filters (frosted glass, yellow, green, blue) is pre-installed. Rotate the slider to select filters for specimen matching, color reproduction, and contrast enhancement.
Polarized Light Observations: Use the polarizer/analyzer set to reveal polymers, dirt, and foreign materials, enhance contrast, and remove glare. Rotate the polarizer to achieve a dark field of view and adjust light intensity. Reduce brightness after observation to prevent visual impairment.
Diagrams (Fig. 16-19) illustrate these procedures.
Darkfield Observation Procedure
For darkfield observations, pull out the brightfield/darkfield knob to position . Pull out the analyzer and polarizer knobs. Open field and aperture diaphragms fully. Place and focus the specimen. If the specimen is not visible, switch to brightfield, focus, then return to darkfield. Adjust collector and lamp focus knobs if asymmetry is observed. Set brightness adjustment to maximum.
Diagram (Fig. 20) shows darkfield controls.
Using Optional Equipment
Eyepiece with a Scale: Used for comparative analysis of linear dimensions. The scale is installed in the eyepiece. A stage micrometer is required for calibration.
Using a Calibration Slide with a Camera: The calibration slide (stage micrometer) is used to calibrate image analysis software. Capture images of the micrometer scale with each objective and use the software's calibration function to establish measurement units.
Diagram (Fig. 21) shows a calibration slide.
