heliX® Adapter Chip Overview
The heliX® Adapter Chip features two distinct spots, each with a different anchor sequence designed for DNA-encoded addressing. Spot 1 is functionalized with an ssDNA template (Adapter strand 1 - nf), while Spot 2 is functionalized with a dsDNA template (cTemplate Adapter strand 2 - nf). These anchor sequences are fixed to the chip surface.

Preparation
IMPORTANT: Both Adapter strands are already pre-hybridized. Adapter strand 1 is pre-hybridized to the Template-32-P-Ra DNA strand, leaving the upper part as ssDNA. cTemplate-Adapter strand 2 is pre-hybridized to the Template-32-P-Ra DNA strand, leaving Spot 2 completely as dsDNA.
To prepare the solution for biochip functionalization, mix the Adapter strand 1 - nf / Template-32-P-Ra DNA (200/250 nM) and the cTemplate Adapter strand 2 - nf / Template-32-P-Ra DNA (200/250 nM) in the same vial at a 1:1 ratio (v/v).

Assay Setup in heliOS
This kit is designed for studying the enzymatic activity of nucleic acid modifying enzymes such as polymerases, reverse transcriptases, and helicases.
To set up an assay, navigate within the heliOS software: go to heliOS > create a New Assay Workflow > add Custom Assay > load Enzyme Binding and Activity. Modify the parameters as needed and run the assay. Suggested assay parameters, including flow rates, functionalization time, and LED power, are available within the heliOS assay setup.
