3.1 Initial Setup

4. Operating Instructions: Performing the Nitrite Test

Follow these steps carefully to ensure accurate nitrite measurements:

  1. Prepare the Sample: Fill the test tube with 5 ml of the pond water to be tested. Ensure the water sample is taken from a representative area of the pond, away from surface film or debris.
  2. Add Reagent 1: Add 5 drops of Nitrite Reagent 1 to the test tube. Cap the test tube and gently invert it several times to mix the solution.
  3. Add Reagent 2: Add 5 drops of Nitrite Reagent 2 to the test tube. Cap the test tube again and gently invert it several times to mix thoroughly.
  4. Wait for Reaction: Allow the test tube to stand undisturbed for 5 minutes. During this time, the color of the solution will develop.
  5. Compare Results: After 5 minutes, compare the color of the solution in the test tube with the color chart provided in the kit. Hold the test tube against the white background of the color chart in good lighting conditions (preferably natural daylight).
  6. Interpret Reading: Match the color in the test tube to the closest color on the chart. The corresponding number indicates the nitrite concentration in mg/L (ppm).

Note: For best accuracy, perform tests at consistent times of day and under similar lighting conditions. Rinse the test tube thoroughly with distilled or deionized water immediately after each use.

5. Maintenance

Proper maintenance of your test kit components will ensure longevity and accuracy:

  • Cleaning Test Tubes: After each test, thoroughly rinse the test tube(s) multiple times with clean tap water, then with distilled or deionized water to remove any residual chemicals. Allow them to air dry completely before storing.
  • Reagent Storage: Store reagents in their original bottles, tightly capped, in a cool, dark place away from direct sunlight and extreme temperatures. Do not refrigerate unless specifically instructed.
  • Expiry Dates: Check the expiry dates on the reagent bottles. Expired reagents may yield inaccurate results and should be replaced.
  • Kit Storage: Store the entire kit in its original packaging in a dry, safe place to protect components from damage and contamination.

6. Troubleshooting

If you encounter issues while using the test kit, consider the following:
