1. Introduction

The MACHEREY-NAGEL Quantofix EDTA test strip is designed for the rapid and reliable determination of EDTA (Ethylenediaminetetraacetic acid), NTA (Nitrilotriacetic acid), and other complexing agents in various solutions. 2. Safety Information

Always handle chemical samples with appropriate personal protective equipment (PPE), such as gloves and eye protection. Avoid direct contact with the test strip reagent pads. Store the test strips in their original packaging, away from direct sunlight, moisture, and extreme temperatures, as specified in the storage instructions. Keep out of reach of children and pets. Dispose of used test strips and samples according to local regulations for chemical waste.

3. Setup and Preparation

  1. Ensure the sample to be tested is at room temperature (15–25 °C).
  2. Open the test strip container only when ready to perform a test to minimize exposure to humidity.
  3. Prepare a clean, dry surface for placing the used test strip.

4. Operating Instructions

Follow these steps for accurate testing:

  1. Remove one Quantofix EDTA test strip from the container. Immediately reseal the container.
  2. Dip the test strip completely into the sample solution for 1 second.
  3. Remove the strip and shake off any excess liquid.
  4. Wait for 15 seconds.
  5. Compare the color of the reaction zone on the test strip with the color scale provided on the test strip container.

5. Interpretation of Results

The color change on the test strip corresponds to the concentration of EDTA or other complexing agents in the sample. Match the color on the strip to the closest color block on the provided color scale. The concentration value indicated next to the matching color block is your result.

For accurate readings, ensure good lighting and compare the strip against a white background if necessary. If the color falls between two blocks, estimate the concentration accordingly.

6. Applications

The Quantofix EDTA test strips are versatile and can be used in various applications:

  • Washing and Cleaning Solutions: Ideal for checking the concentration of complexing agents like EDTA and NTA, which have replaced phosphates as additives in these solutions.
  • Proteomics Laboratories: Used to control if EDTA has been completely rinsed out after regenerating Ni- and Co-precharged chromatography columns (FPLC columns) for purifying recombinant proteins. This can significantly reduce rinse time and increase throughput.

7. Maintenance, Storage, and Disposal

Storage:

  • Store the test strips in the tightly closed original container at room temperature (15–25 °C).
  • Protect from moisture, direct sunlight, and heat.
  • Do not refrigerate or freeze.

Disposal:

Used test strips and samples should be disposed of in accordance with all applicable local, state, and federal regulations for chemical waste. Consult your facility's waste management guidelines.

8. Troubleshooting

  • No Color Change or Faint Color: Ensure the test strip was fully immersed in the sample. Check the sample for extreme pH values that might interfere with the reaction. Verify the test strip's expiration date.
  • Inconsistent Results: Ensure consistent dipping time (1 second) and reaction time (15 seconds). Avoid touching the reagent pad with fingers. Use fresh samples and strips.
  • Expired Strips: Do not use test strips past their expiration date, as accuracy cannot be guaranteed.
