To prepare your cell samples for analysis using the Cellaca MX, follow these steps as outlined in the user manual:
1. **Inverting and Mixing Solutions:** Invert the tube containing cells or counting beads ten times to ensure homogeneity. Vortex bead solution for 10 seconds if using counting beads. Do not vortex cell samples.
2. **Loading Sample:** Load 50 μL of bead solution or cell sample into the Loading Well of the counting plate. If using a cell sample that needs to be mixed, use the Mixing Well of the counting plate to prepare the sample before loading it into the Loading Well.
3. **Plate Placement:** Place the prepared plate onto the instrument stage, aligning the notched corners of the plate with the stage accordingly (e.g., well A1 is in the top left corner).
4. **Setup Details:** Enter a Plate Name, select an appropriate assay from the drop-down menu, and enter a Dilution Factor in the Setup Details area of the software.
5. **Well Selection:** Ensure the displayed Plate Type is correct, then click on the individual wells used to load the sample. You can select or clear multiple wells by clicking and dragging your mouse or by using the Select All button.
6. **Previewing the Sample:** Click the Preview button to view the sample. Adjust the focus and verify fluorescent exposure as needed.
Remember that cell concentrations of 1.0 x 105 to 1.0 x 107 cells/mL can be analyzed on the Cellaca MX, with a concentration of 1.0 x 106 cells/mL being optimal. Avoid shaking or vortexing the sample as this may damage cell membranes.
For additional details and specific guidelines on preparing cell samples for analysis with the Cellaca MX, refer to the Cellaca MX User Manual provided with the instrument.
