Setup
- Unpacking: Carefully remove all components from the packaging. Retain the packaging for future transport or storage.
- Placement: Place the microscope on a stable, vibration-free surface away from direct sunlight and heat sources.
- Head Installation: Loosen the head retaining screw on the top of the microscope body. Gently insert the binocular head into the opening, ensuring it is fully seated. Tighten the retaining screw.
- Eyepiece Installation: Remove the protective caps from the eyepiece tubes. Insert the desired eyepieces (10X or 25X) into the tubes.
- Objective Lens Installation: Rotate the revolving nosepiece to ensure there is no objective lens directly above the stage. Screw the objective lenses into the nosepiece in increasing order of magnification (e.g., 4X, 10X, 40X, 100X).
- Power Connection: Connect the power cord to the microscope's power input and then to a suitable electrical outlet.
- Digital Eyepiece Camera Installation: If using the digital eyepiece, remove one of the standard eyepieces and insert the 1MP digital eyepiece camera into the eyepiece tube. Connect the camera to your computer via USB.
Operating Instructions
1. Brightfield Observation

- Power On: Turn on the main power switch. Adjust the LED illuminator brightness using the dimmer control.
- Stage Adjustment: Lower the mechanical stage using the coarse focus knob to create enough space to place a slide.
- Slide Placement: Place a prepared specimen slide onto the mechanical stage, securing it with the stage clips. Use the X-Y stage controls to position the specimen over the light aperture.
- Magnification Selection: Rotate the revolving nosepiece to select the lowest power objective lens (e.g., 4X). Ensure it clicks into place.
- Focusing:
- While looking through the eyepieces, slowly raise the stage using the coarse focus knob until the specimen comes into rough focus.
- Use the fine focus knob to achieve sharp focus.
- Adjust the interpupillary distance (distance between eyepieces) and diopter adjustment on the eyepiece for comfortable viewing and to correct for differences in vision between your eyes.
- Increasing Magnification: Once focused at low power, rotate the nosepiece to a higher power objective (e.g., 10X, 40X). The microscope is parfocal, meaning only minor adjustments with the fine focus knob should be needed.
- Using 100X Oil Immersion Objective:
- After focusing with the 40X objective, rotate the nosepiece halfway between the 40X and 100X objectives.
- Place a small drop of immersion oil directly onto the center of the specimen slide.
- Rotate the 100X objective into the oil drop. The objective lens should make contact with the oil.
- Use only the fine focus knob to bring the image into sharp focus. Do not use the coarse focus knob with the 100X objective.
- After use, clean the 100X objective lens and the slide immediately with lens cleaning solution and lens paper.
- Condenser Adjustment: Adjust the iris diaphragm on the condenser to control the amount of light and contrast. For higher magnifications, open the diaphragm more. For lower magnifications, close it slightly. The condenser can also be centered for optimal illumination.
2. Darkfield Observation
The AmScope B120C-DK-E1 includes a darkfield condenser for observing unstained, live specimens.
- Condenser Exchange: Remove the brightfield condenser from its mount beneath the stage. Carefully insert the darkfield condenser into the mount. Ensure it is seated correctly.
- Specimen Preparation: Darkfield observation works best with thin, unstained specimens.
- Illumination: Turn on the LED illuminator and set it to a high brightness level.
- Focusing: Use a low power objective (e.g., 10X or 20X, if available) first. Focus on the specimen as you would in brightfield. The specimen will appear brightly lit against a dark background.
- Centering the Darkfield Condenser: Some darkfield condensers require centering. If your condenser has centering screws, adjust them until the darkfield effect is uniform across the field of view.
- Higher Magnification: For higher magnifications, you may need to use a specialized darkfield objective or ensure your standard objectives are compatible. Always ensure sufficient immersion oil is used with 100X darkfield objectives.
3. Digital Eyepiece Camera Operation
The included 1MP digital eyepiece camera allows for image capture and live viewing on a computer.

- Software Installation: Install the professional microscopy software provided (or download the latest version from the AmScope website) onto your Windows, Mac, or Linux computer.
- Connect Camera: Insert the digital eyepiece camera into one of the microscope's eyepiece tubes and connect it to your computer via the USB cable.
- Launch Software: Open the installed microscopy software. The software should detect the camera automatically.
- Live View: The software will display a live feed from the microscope. Adjust microscope focus and illumination for the best image.
- Image/Video Capture: Use the software's functions to capture still images or record videos of your observations. The software also provides tools for image processing and measurement.
Maintenance
- Cleaning Lenses: Use a soft brush to remove dust from lens surfaces. For smudges or oil, use a small amount of lens cleaning solution on lens paper. Wipe gently in a circular motion. Never use abrasive cloths or harsh chemicals.
- Cleaning Body: Wipe the microscope body with a soft, damp cloth. Avoid getting moisture into electrical components.
- Storage: When not in use, cover the microscope with a dust cover to protect it from dust and debris. Store in a cool, dry place.
- Immersion Oil: Always clean immersion oil from the 100X objective and slides immediately after use. Dried oil can be difficult to remove and can damage the lens.
